We just moved in and there has been no landscaping done in the back yard. I'd like to start seeding ASAP, but right now there are tons of weeds. My question is, what is the best way to remove the weeds before laying new soil and grass?

Should I pull them by hand, then spray a weed killer then rototill?

Personally, I would never, ever spray poison in my yard. You might get rid of the plants you don't want right now, and later discover that the plants you do want can't grow there either. I would probably till as deeply as possible (hand digging and turning is better, but hard work) then rake and pull out the weeds once they're loosened. That soil looks hard and nutrient-poor, so it probably needs a lot of remediation before you sow or sod. Can't tell by looking what it needs.

I would use a systemic weed or vegetation killer but you cannot be in a hurry as they have to be dead, totally but first pull the largest ones.
If you roto-till to soon you will simply be spreading seeds they may have dropped and they WILL come back.
Find out what weeds you actually have and it is best to get a dedicated weed killer for them.

A total vegetation killer would, hopefully kill all, but after years of using many brands including much ballyhooed Roundup, they can be , too often , under whelming.

Spray the area with Roundup!! Then within 24 hours you can put out seed or
sod. If you do seed, get a good perennial rye. Do it sooner than later for your
area. Then in the spring toss more out as you want over seed for a good carpet
of grass.

I have an neighbor that just redid their yard, scraped and tilled the whole front yard,
did spay at all and now they are dealing with weeds.
